J&J’s abiraterone shows survival improvement by about four months
The results from a Phase III clinical trial demonstrated that an oral androgen synthesized inhibitor by Johnson & Johnson, abiraterone acetate, improves the survival rate by about 4 months if compared with the placebo effect in patients suffering from prostate cancer aggravated by visceral metastases.
The interim analysis of the COU-AA-301 study of 1,195 patients presented at the 35th annual congress of European Society for Medical Oncology (ESMO) in Milan, Italy, showed a great survival rate and good safety profile of abiraterone.
The company revealed its COU-AA-301 clinical trial along with the recommendations of its Independent Data Monitoring Committee, which suggested that the patients taking placebo should be also treated with abiraterone acetate combined with low doses of prednisone.
Originally designed by Prof. Mike Jarman and other personnel of the Cancer Research Therapeutics Unit at Britain’s Institute of Cancer Research (ICR) and funded by Dr. Lindsay Rosenwald and his company, abiraterone is going to be filed for approval in the US and Europe by the end of 2010. Being one of the leading investors in the biotechnology sector, Dr. Rosenwald and his portfolio companies promote the development of promising discoveries and their commercialization. As one of the most promising inventions for the treatment of prostate cancer, abiraterone will be applied by Johnson & Johnson for approval in the rest of the world according to local regulations.
Once approved, abiraterone acetate will be distributed by Lindsay Rosenwald’s portfolio company, Centocor Ortho Biotech Inc, in the US. The rest of the world will see the drug thanks to Janssen Pharmaceutical Companies.
In 2009, abiraterone was acquired along with Lindsay Rosenwald’s portfolio company, Cougar Biotechnology, for US $ 970 million, which in its turn had licensed abiraterone acetate from the BTG life sciences company in the UK.
Improved survival in 36%
The COU-AA-301 trial Phase III was made in 13 countries and included patients suffering from prostate cancer aggravated by visceral metastases. A total of 797 patients took abiraterone acetate (in the dose of 1,000 mg a day) in a combination with prednisone or prednisolone (5 mg twice a day) and 398 patients took placebo with prednisone or prednisolone. The interim trial demonstrated that the patients treated with abiraterone acetate were less prone to fatal outcome, while the placebo group had a 36% increase in death rate. The prostate-specific antigen (PSA) progression time increased from 6.6 months to 10.2 months in the placebo group, while the abiraterone patients had significant improvements.
38% of patients undergoing the abiraterone treatment achieved total PSA response, while only 10% achieved the same in the prednisone and placebo group.
According to Dr. Lindsay Rosenwald and Dr. Johann de Bono, who were among the lead COU-AA-301 investigators, abiraterone acetate has all the potential to meet the needs of the biomedical sector and is extremely important to the patients with prostate cancer as well as their families.
The results were very promising because men with metastatic advanced prostate cancer have very few treatment options and poor results.
New treatment possibilities
Andrew Paramore, one of the leading Datamonitor analysts, believes that the results of the abiraterone treatment are striking, since very few drugs have been as effective as abiraterone acetate. However, Paramore says that choosing the drug for individual patients may be “a difficult task as two other drugs, Jevtana (carbazitaxel; Sanofi-Aventis) and Provenge (sipuleucel-T; Dendreon), have also shown a survival advantage, albeit in different subsets of patients”.
Now there are two possible treatment directions according to Paramore: the combination of the new drugs with each other or with other existing and already approved drugs. This will be done to increase the possible survival benefits as well as to improve the quality of life in patients with prostate cancer.
